Cuenca lies on the Pan-American highway. Buses offer connections to many cities in Ecuador. Central Cuenca is easily walkable, and it is often faster than taking a cab through the narrow traffic-jammed lanes. Cabs are readily available and charge $1-2 ($1 more at night) per trip. The fee should be negotiated before entering the car.
